“It’s all about knowing your nanoparticles”: characterization as a scientific and regulatory problem

Characterizing nanoparticles is a complex problem that combines scientific and regulatory issues. From a research perspective, it increases the reproducibility of experimental results, a problem that is not specific to nanomedicine but that has been raised for all biomedical research. Notably, in a survey of 1,576 scientists conducted by the journal Nature, 70% of respondents failed to replicate the experiments of another team, and 50% failed to replicate their own experiments. From the same sample, 90% of respondents working in chemistry and 80% of respondents in biology said they had failed to reproduce other teams’ experiments at least once (Baker, 2016). The debate on the “reproducibility crisis” in biomedical research has also involved pharmaceutical and biotechnology companies (such as Amgen in the United States and Bayer in Germany), who failed to reproduce scientific results they had intended to use for clinical developments (Begley & Ellis, 2012). Nanomedicine researchers identified the poor characterization of nanoparticles as the main obstacle to the reproducibility of experiments:

One can reproduce/replicate something only when it is known precisely what is to be reproduced. At this stage, the knowledge on most of the nanoparticle drug formulations is far from adequate and the old question “What’s in the bottle?” continues to frustrate researchers.
